Hitting a cucco too many times in Ocarina of Time. Nothing like being chased around (and possibly killed) by a flock of vengeful chickens. XD
Characters bantering while you run around in White Knight Chronicles. Caesar makes me laugh cause he says the most random stuff sometimes. Battle commentary. "Do I look badass or what?"
Skits in Tales of the Abyss. Anything involving Jade.
Pulling hate from a boss mob in Final Fantasy XI just long enough to get one-shotted for a ridiculous amount by its 2-hour. Tonberry rancor. Bards.

Yep definitely one of my favorites, I always play as a Dragoon and let's just say my Dragoon was too much of a beast lol, I once got one shotted for over 6000 lol, one of the new abyssea mobs knife me while he was invisible and all I saw was numbers lmao....
Legendary moment, I still need to pay him back as well >.>

The Metal Gear Solid games had many things that amused me, haha. There's one in particular in Snake Eater when you find yourself in a cave with Eva, and depending on how things went before you got there, she'd fix you up. At this point the camera aims at the wall so you see the shadows, and it just looks wrong. So funny though. xD
The whole Wall Market part of Final Fantasy VII amused me, mainly because I'd played through it when I was younger and didn't quite understand what was going on - when I was older, I understood why my mum had made a couple of "WTF?" faces when she watched me play. :D
The Sims games never fail to amuse me. I get creative with just about everything, so I've built some really cool houses and crap. The latest thing that had me almost crying with laughter was watching my pregnant sim dance with her husband. I then went on a hunt for another pregnant sim, because I'd remembered seeing one in town, and I invited her over for a dance and found that even more funny. ^^;
LittleBIGPlanet... well, it's hard to have difficulty not amusing yourself in that one. Especially if you're with friends. Sure, dragging your friends around and slapping them about (and vice versa) gets dull after a while, but it's funny.
Finally, Dragon Age. Just talking with Alistair makes my heart light up - the stuff he comes out with! WHY CAN'T THERE BE PEOPLE LIKE HIM IN REAL LIFE?! 8-():love::o

When the main character is oblivious to an insanely dangerous and obvious plot twist, walks into the fight completely unprepared, and wins
When you kill a boss with your weakest attack which you used accidentally.
In arc rise fantasia a couple of things: rydia being so oblivious, l'arc being so kind and easily flustered despite being extremely similar to cloud, leslie's flirting, and cecille beating the crap out of niko with a single punch

I'd have to say moments like those in the Metal Gear Solid series: Raiden gets a sex check from the President (I believe it's the President). Self-aware moments: one villain is named Vamp within the context of a serious spy-espionage-military-thriller that is portrayed in very realistic ways.
To have characters named Vamp, Fortune, etc. within such a context makes me laugh because it's pushing the boundary between merely being a game and saying, "Hey, I'm aware I'm a game. Moreover, I'm aware that you're aware that I'm aware you're a game." Similar to the previous statement is the existence of a psychic named Psycho Mantis mentioned, again, like many other characters in the Metal Gear Solid, in a seemingly serious, realistic gaming context.
Any time Snake is insulted in Metal Gear Solid 4 and his psyche goes down; on the other hand, any time Snake's psyche meter goes up due to ogling at a woman or some other funny reason.
